Attempting to shoot downtown sidewalk areas and intersections. Patience is required, but beyond that, can masking be used to remove moving objects (people, cars) that come into frame?

yes, absolutely. They will cover some areas obviously, so i suggest taking a few extra pictures so that if a car comes into frame and you have to mask it, the next photo(s) will hopefully not have it.
You can use 3DF Masquerade (suggested) or mask it with other tools if you wish to. -
